【求助】IKK-NF-κB通路抑制剂NBD

你好,本人要做IKK-NF-κB通路方面的研究,做的是胃上皮细胞,指导导师让我用NBD抑制IKK活性,并要求我订购IKKβ磷酸化抗体做IKK活性试验,我有点困惑,查了不少的文献,IKK都是引起IκB磷酸化激活NF-κB的,而非IKK磷酸化激活的,其中的原理是什么?NBD多肽我在文献上看到有野生型,突变型,好多文献里NBD多肽是找公司合成的,肽链有长有短,打电话问公司订,他们要求我提供序列,我也不知道提供什么?恳请各位大侠给出高见,谢谢。

原理如下:
IKKγ binds to specific sequences on IKKα and IKKβ, forming the IKK complex. The IKK complex phosphorylates downstream signaling molecules during NF-κB activation. The IKKγ inhibitory peptide contains an IKKα and IKKβ consensus binding sequence, called NEMO binding domain or NBD. When IKKα and IKKβ are bound to the IKKγ NBD peptide, they can not bind to IKKγ. Thus the formation and function of the IKK complex is blocked.

谢谢,我找到买到NBD多肽的网站了,导师让我订购IκB 磷酸化抗体来测定是否抑制IKK活性,我不知道抑制后IκB 磷酸化表达增强还是减弱,因为IκB活化后会被磷酸化再被26S蛋白小体降解,而没被活化的IκB则不会被磷酸化。
Absolutely, when you use NBD to inhibit the IKK complex, the P-IkBa will be inhibited. But, you need to stimulate the NFkB pathway after you add NBD.
