为克隆EGFP(模板为pEGFP-N2),我设计了一对引物:
P1: 5' AGCTCAAGCTTCGAATTCTGCAGTC 3' 有酶切位点EcoR I.(GAATTC)
P2: 5' CTCGGATCCTACAAATGTGGTATGG 3' 有酶切位点BamH I(GGATTC)
用primer5.0 设计,rating 为79分,但两个引物都有Dimer,且P1 中Dimer的自由能高达-11.2,共有四条-10---- -11.2的Dimer.
各位给看看吧,谢谢.这样的引物能扩增出800多的片段吗?能否讲讲,急啊!!
