Antibody Epitope Mapping Using Yeast Display

Many approaches for antibody epitope mapping utilize peptides or peptide fragments and are limited to antibodies binding linear epitopes or epitopes not requiring fully folded conformationally correct antigens. The protocols in this chapter show how yeast displayed antigens and antigen libraries can be used to map antibodies gross and fine epitopes down to the functionally important amino acids. Methods are also described to measure the impact of individual antigen amino acids on the energy of antibody binding.
