Purification of exosomesWelton et al., have purified exosomes from the culture medium of a bladder cancer cell line HT1376 . After removing the cells by a gentle spin (400 g for 10 min) and the cellular debris (2000 g for 15 min and then 10,000 g for 30 min), the supernatant was subjected to high speed centrifugation (100,000 g for 2h) by laying on top of a 30% sucrose cushion in deuterium oxide (D2O or heavy water). After a wash with PBS, the pellet contained the exosomes. Hogan et al., have isolated exosomes from urine with the following method [9]. The urine sample was precleared by a 15,000 g centrifugation and passed through an 8 μm nylon filter before being re-centrifuged at 150,000 g for 1h. The pellet contained exosome-like vesicle and Tamm-Horsfall protein (THP; or uromodulin), the main protein in urine . To remove THP, the sample was layered on top of a discontinuous sucrose gradient (5-30%) in D2O with pH 6.0. After centrifugation at 200,000 g for 24h, the cushion was collected in 14 equal fractions. Each fraction was diluted 5- to 10-fold in PBS and re-centrifuged at 150,000 g for 1h and the pellets with pure exosomes were recovered.
