POLY-L-LYSINE COATED SLIDES
SOLUTIONS
--5% Chromic acid
--0.01% Poly-L-Lysine (PLL) - made from a dry protein, with distilled water. (100mg/1 litre)
This procedure can be carried out on either a ski or carousel staining machine.
SKI STAINING MACHINE
1. Remove the new slides from box.
2. Clip them individually onto the ski using the correct clips x25 slides maximum.
3. The procedure takes approximately 20 minutes, and the slides pass through the following solutions in this order.
i) Chromium trioxide (chromic acid 5%) x6
ii) Distilled water x6
iii) P-L-L solution x1 (double)
iv) Absolute methanol x2
4. The slides come off the machine dry and should then be put into an empty slide box.
5. The box should then be labelled 'P-L-L' and dated.
6. The slides are then ready for use.
SHANDON ELLIOT CAROUSEL
1. Remove the new slides from box.
2. Place slides in the upright slide rack. Approximately 25 slides.
3. Hook the slide rack into the carousel, above the first solution bath (chromic acid).
4. Set the dial on the face of the carousel to the 'start' position.
5. Start the machine.
6. The procedure takes approximately 35 minutes and the slides pass through the following solutions in this order.
i) 5% chromic acid x1
ii) Distilled water x5
iii) P-L-L solution x1
7. Dry the slides in the rack in the 37°C oven for 24 hours.
8. Put the slides in the rack into the empty box.
9. The box should be labelled 'P-L-L' and dated.
10. The slides are then ready for use.
