急问，怎样才能使前体miRNA 主要表达 star miRNA (miRNundefined)
在下正在作star miRNA 的研究，想把前体miRNA 克隆到载体，然后表达成熟miRNA，但有一个问题，怎样才能使前体miRNA 主要产物为star miRNA (miRNundefined)呢，或者说怎样才能构建主要表达star miRNA (miRNundefined)的克隆呢，先谢谢了

My suggestion is using antisense inhibitor to block the dominant counter-mir from your pri-miRNA expression. But by doing this, I still don't think the miundefined is "OVEREXPRESSED" (if this is what you want?), the miundefinedjust becomes more dominant. The similar way is to change the C on the counter-mir to U (T) to eliminate its function but keep the base-pair.

You can also try to make siRNA-like double-stranded RNA with GC base pairing in one end to favor the miRNA machinery to take the miundefined sequence, however, DICER might chop your "siRNA" into 19-mer and I think that's the drawback. Alternatively, if you can change the base-pairing in the pre-miRNA to make miundefined being favored, then it might work.

个人意见：直接合成你要表达的片段，接到过表达启动子上。protocol online上好像有这样的做法。

人工构建前体，如miR393undefined：miR393undefined，在这样的前体中miR393undefined即成为成熟体过量表达，而miR393undefined被降解或不起作用，（miR393undefined即miR393b）

请问楼上的miR393undefined是如何设计的？？我也想用质粒只表达一个链。
