Strategies for Regulating Calpain Activities in Living Cells

The goals of our research program have been to understand the mecha-nism(s) responsible for the turnover of myofibrillar proteins of skeletal muscle. This understanding would be useful in development of strategies to treat muscle wasting diseases and, possibly, to augment efficiency of muscle growth in domestic animals. The challenge in this area is that several proteolytic systems have been implicated in this process. For example, Goll et al. (1 ,2 ) have summarized in considerable detail a logical rationale for the participation of the calpains in this process. However, other researchers (3 5 ) have provided compelling evidence that the proteasome, the large, multifunctional ATP-depen-dent protease, plays a key role in this process as well. Earlier studies (6 ) have also documented the participation of the lysosome in this process.
