PCR扩增出非特异带但无特异带,是不是引物的特异性不好?

欲通过RT-PCR检测一种基因的不同剪接异构体表达情况,
在文献(cancer research, oncogene)中查到的两对引物序列如下:
P1 5' atg agg atc cca tgg gcc agc a 3'
P2 5' aag ggc ttt tca ctt gtt tta c 3'
P1 5' atg agg atc cca tgg gcc agc a 3'
P3 5' aag ggc ttt tca cct gta tga g 3'
RT用的是MBI的试剂盒,反应体系20ul:总RNA 4.0 ug, oligo 1 ul,M-Mulv 1 ul,反应条件为70℃5分钟,37℃5分钟,42℃60分钟,70℃10分钟.
PCR体系25ul:
cDNA 5ul
Mgcl2 1.0mM
dNTP 200uM
引物 0.2uM
TaQ酶 1U
PCR条件94℃1min---51℃1min(还试过55℃1min) ---72℃ 1min---35cycles----72℃ 5min. (文献中退火温度是个大致范围50-60℃)
结果出现了两三条非特异性条带,郁闷的是均并没有预计大小的特异性扩增带,不知是引物有问题,还是需要进一步改变反应条件?急盼各位高手帮忙分析一下引物及反应条件,并请指教该如何改动。不胜感激!
