NMR spectroscopy is an extremely valuable and versatile method for studies of nucleosides, nucleotides, and their oligomers. It can provide information about chemical composition, three-dimensional structures, internal motions, and interactions with other macromolecules or ligands. It can be used to follow outcome and yield of a synthesis, to ascertain identity and purity of the product, to identify and quantify byproducts, and finally, to obtain a largely complete picture of the spatial arrangement of atoms in the molecule. It can be used at all stages of synthesis, development, and investigations of oligonucleotides.
