BAPTA是由Tsien博士发明的一种钙选择性螯合剂。(logKCa=6.97,logKMg=1.77)它基本的螯合单位和EDTA类似,只是两个脂肪氮被芳香氮所替代。因此,BAPTA在生理pH下不会被质子化。BAPTA的pKa3为5.47,pKa4为6.36。这个特点显示了去质子化的步骤并不包含在Ca的螯合步骤中。由于不受质子的干扰,它的螯合率比EGTA要高很多。BAPTA-AM是BAPTA的乙酰甲酯衍生物,通过AM法,能被轻易负载到细胞中。BAPTA-AM对于细胞内钙离子的监控是非常有用的。
葡萄糖调节蛋白(glucose regulated proteins, GRPs)是内质网中一类重要的驻留蛋白,包括GRP78和GRP94。 GRP94是热休克蛋白HSP90家族的成员之一,与HSP90具有50%的同源性,是高度保守的蛋白。

Extracellular stress proteinsincluding heat shock proteins (Hsp) and glucose regulated proteins (Grp) are emerging as important mediators of intercellular signaling and transport. Release of such proteins from cells is triggered by physical trauma and behavioral stress as well as exposure to immunological "danger signals". Stress protein release occurs both through physiological secretion mechanisms and during cell death by necrosis. After release into the extracellular fluid, Hsp or Grp may then bind to the surfaces of adjacent cells and initiate signal transduction cascades as well as the transport of cargo molecules such as antigenic peptides. In addition Hsp60 and hsp70 are able to enter the bloodstream and may possess the ability to act at distant sites in the body. Many of the effects of extracellular stress proteins are mediated through cell surface receptors. Such receptors include Toll Like Receptors 2 and 4, CD40, CD91, CCR5 and members of the scavenger receptor family such as LOX-1 and SREC-1. The possession of a wide range of receptors for the Hsp and Grp family permits binding to a diverse range of cells and the performance of complex multicellular functions particularly in immune cells and neurones.

It has been established that as molecular chaperones, the glucose-regulated proteins (GRPs) play an important role in maintaining cellular homeostasis. This conventional concept of GRPs as protein folding chaperones is updated by discoveries that GRPs promote tumor proliferation, metastasis, drug resistance, immunotherapy, which have major clinical implications in the prognosis and treatment of cancer. Further, the localization of GRPs on the cell surface of certain cell types suggests that they serve new functions as cell surface receptors for signaling. These and other new developments on the role of GRP78, GRP94 and GRP170 in cancer progression and therapy are discussed in this review.

Endoplasmic reticulum (ER)-resident stress proteins in particular are interesting model proteins as they perform crucial functions in an organelle that responds promptly to cell stress. We describe transcriptional regulation of ER-resident stress proteins, their involvement in the cellular response to infection and discuss their potential as vaccine candidates against infectious diseases.

Heat shock proteins (HSPs) are a large family of proteins with different molecular weights and different intracellular localizations. These proteins undertake crucial functions in maintaining cell homeostasis, and therefore they have been conserved during evolution. Hsp70 and Grp94/gp96, due to their peptide chaperone capacity and their ability to actively interact with professional antigen-presenting cells (APCs), are also endowed with crucial immunological functions. The immunological properties of these proteins and their implications for vaccine in human cancer will be discussed. Immunological and clinical data of phase I/II studies in melanoma and colorectal cancer patients will be reviewed.
