Hemoglobin (Hb) is an important oxygen transport protein mainly present in the red blood cells circulating in the blood. Measuring brain Hb content has been used to assess hematoma mass after intracerebral hemorrhage. Recent studies have demonstrated that Hb is also present in the neurons of embryonic and adult brains. In this chapter, we introduce two methods for quantitation of Hb levels in the brain, including spectrophotometric assay and enzyme-linked immunosorbent assay (ELISA). The details of the procedures, along with results calculation, advantages, and limitations are discussed.
