Hi,Can you tell me what do "SSC" in a southern blot, how he act on DNA or in the protocole.

(excuse-me for my traduction, i never write in english)

"SSC" is a abbreviation of sodium chloride and sodium citrate solution, which used to transfer nuclear acid from gel to membrane ( 20 *SSC or 1undefinedSSC or undefinedSSC with alkaline). In Roche's DIG system, they said that the high solt concentration SSC shows much efficiently than the others like valcuum or electrophoresis.

DNA is transferred most efficiently to nitrocellulose as a sodium salt where 90% of the effective negative charge has been neutralized by sodium chloride/citrate. Good Luck.
