CHO-K1 (hamster, Chinese, ovary)

ECACC 85051005

Morphology: Epithelial

Hamster Chinese ovary

Depositor: Dr D Newell, PHLS CAMR, Porton Down, Salisbury, UK

No restrictions. Patent: None Specified By Depositor

Properties: Applications: Nutritional and gene expression studies

Available in the following LABORATORY:

CAMR Centre for Applied Microbiology & Research (ECACC, Salisbury, Wiltshire)

Ham's F12 + 2mM Glutamine + 10% Fetal Bovine Serum (FBS). Split confluent cultures 1:4 to 1:10 i.e. seeding at 1-2x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37C. Passages: 17 Karyotype: Hypodiploid, modal No 20

Hazard: CY

A subclone of the parental CHO cell line, which was derived from the ovary of an adult Chinese hamster. Cells require proline due to the absence of the gene for proline synthesis, the block in the biosynthetic chain lies in the step converting glutamic acid to glutamine gamma serialdehyde. They undergo morphological changes in response to cholera toxin.

CHO-K1 (hamster, Chinese, ovary)

DSMZ ACC 110

Morphology: adherent, fibroblastoid cells, may be induced to undergo additional differentiation

Chinese hamster ovary cells subclone from parental CHO cell line that was initiated from an ovary biopsy of an adult Chinese hamster in 1957 confirmed as hamster with IEF of NP, PEP B

Viruses: ELISA: reverse transcriptase negative

Depositor: Dr. P. E. Bryant, University of St. Andrews, St. Andrews, Fife, UK

General Restrictions

Available in the following LABORATORY:

German Collection of Microorganisms and Cell Cultures (DSMZ, Braunschweig)

90% Ham's F12 + 10% FBS split confluent cultures 1:3 to 1:10 using trypsin/EDTA; seed out about 10 5 - 10 6 cells in an 80 cm 2 flask; cells grow up in 3-4 days to confluence at 37C with 5% CO2 about 10 x 106 cells/80 cm 2 or 3 x 10 6 cells/25 cm 2; doubling time of ca. 24 hours

Further information:

frozen with 70% medium, 20% FBS, 10% DMSO at about 1-2 x 106 cells/ampoule; negative in DAPI, microbiological culture, RNA hybridization, PCR assays

Fingerprint: same Hinf I-(gtg) 5 DNA profile as CHO-DHFR

Cytogenetics: near diploid karyotype with 12% polyploidy; 22(20-23); exhibits distinctive CHO karyology
