Protocol for Construction of BAC Libraries
The bacterial artificial chromosome cloning (BAC) system is emerging as the system of choice for construction of plant genomic libraries with average inserts of 150 kb. The intent of the workshop is to provide detailed training in the theory and practice of construction, characterization and screening of plant BAC libraries including the use of robotics. Participants will use their own plant materials and at the end of the workshop return to their laboratory with megabase-size DNA suitable for the generation of a BAC library with large inserts.
The protocol consists of methods that have been used successfully by our group to isolate and clone large DNA fragments in bacterial artificial chromosomes.
