剂量效应曲线的累积正态分布拟合

摘要 目的: 研究S型剂量效应曲线的累积正态分布函数拟合。方法: 构建累积正态分布函数的适当表达形式,对作者实验室所获得的实例数据进行拟合,同时与常用的四参数方程拟合进行比较。结果: 两种拟合方法具有高度的一致性。结论: 正态分布模型是一种传统的数学模型,在此处的应用为实验数据拟合提供了另一个可能的选择,并为药效学S型剂量效应模型的研究分析提供了另一种思路。特别地,在常用于药效学S型剂量效应曲线拟合的数学模型中,其Hill系数被认为没有任何生理学含义,而累积正态分布函数中的相应参数却有明确的生理学含义,从数学上体现了药理与药效的内在联系。

Fitting Dose-Effect Curve with the Accumulation Normal Distribution

Abstract OBJECTIVE: Study the fitting about S model dose-effect curve with the accumulation normal distribution function. METHODS: Construct proper accumulation normal distribution function form and fit with our experimental data, then compare with the common 4-parameter equation fitting. RESULTS: The two fitting methods show high uniformity. CONCLUSION: Use accumulation normal distribution, a kind of traditional mathematical model, we provide a possibility for fitting experimental data, and also provide another way for pharmacodynamics S model dose-effect analysis. Especially, the Hill coefficient is considered not any physiological meanings in the mathematics model usually used in fitting pharmacodynamics S dose-effect curve, but the analogous parameter in the accumulation normal distribution function has definite physiological meanings, and indicate the relationship between pharmacology and pharmacodynamics.
