胞质溶胶中合成的蛋白质进入到内质网、线粒体、叶绿体和过氧化物酶体则是通过一种跨膜机制进行定位的，需要膜上运输蛋白(protein translocators)的帮助。被运输的蛋白通常是未折叠的状态，细菌的质膜上也有类似的运输蛋白。
