The isolation of RNA and its subsequent characterization by various applications, such as the generation of cDNA libraries, Northern blot analysis, and reverse transcriptase polymerase chain reaction studies (RT-PCR), are fundamental for understanding the mechanisms underlying gene expression. In order to obtain starting material of high quality, it is crucial to isolate pure and intact RNA from living cells. Effective cellular lysis and membrane disruption, inhibition of endogenous ribonuclease activity, deproteinization of the lysate, recovery of intact RNA, and storage of RNA are the most important factors to be considered when preparing RNA.
