简介

根据本方案制备的哺乳动物 DNA 约 20~50 kb，适于作 PCR 反应的模板。DNA 产量在 0.5 到 3.0 μg/mg 组织之间变化，或者是 5~15 μg/300μl 全血。本实验来源「分子克隆实验指南第三版」黄培堂等译。

操作方法

一、材料 1. 缓冲液及溶液细胞裂解缓冲液（10 mmol/L Tris-Cl (pH 8.0)，1 mmol/L EDTA ( pH 8.0)，0.1%（m/V） SDS）乙醇异丙醇醋酸钾溶液（60 ml 的 5 mol/L 醋酸钾，11.5 ml 冰醋酸，28.5 ml 水）红细胞裂解缓冲液（20 mmol/L Tris-Cl (pH 7.6) TE ( pH 7.6)
